RM Synthesizer:
The preparation of solutions of encapsulated proteins requires fine and reproducible control over pressurized fluids. Daedalus is pleased to introduce the RM Synthesizer; our introductory machine designed to bring this capability into the laboratory setting. It is an intuitive platform that combines all the steps starting with making the sample and finishing with cleaning the chamber after the preparation is complete. Though manually controlled it is more that suitable for the preparation of high quality reverse micelle samples in liquid ethane.

One of the critical issues with handling samples at high pressure is moving the prepared sample from the mixing chamber into the sample tube while keeping the pressure above the encapsulation threshold. This is accomplished by using a pressure driven piston to rapidly displace the sample into the tube thus ensuring maximum sample integrity. An integrated gas booster provides this force and does so without consuming expensive deuterated alkane reagents. This provides the flexibility to load the syringe pump with deuterated ethane for maximum NMR performance without concern for losses from performing support functions with the syringe pump.

The RM Synthesizer requires an Xtreme-10 for delivering the pressurized ethane the device plus the operational gases, ethane, nitrogen, and carbon dioxide. The cleaning manifold bottles and connection hardware is included as is the regulators for nitrogen and carbon dioxide gas cylinders.

Xtreme-10 Syringe Pump:

The Xtreme-10 Syringe Pump is the large capacity model of the Xtreme Syringe Pump line. It is intented for use with the RM Synthesizer for the preparation of samples in liquid ethane. It can deliver fluids at pressures up to 10,000 psi (690 bar). The active volume of the pump is 34.7 ml so the pump can be filled with ethane gas at cylinder pressures and compressed to provide sufficient liquid ethane for sample preparation.
The pressure can be controlled by changing the setpoint or programming the controller to deliver pressure at specific steps during the sample preparation.
The Xtreme works with both liquids and gases.

Reverse Micelle NMR Manifolds and Tubes 1 kbar Rated:
Self-contained cell (1 kbar rated):
This NMR cell and tube combination was designed specifically to address the needs of reverse micelle NMR applications. The tube itself is 3.6 mm I.D. x 5 mm O.D. It has a sample volume equal to approximately 2/3rds of a standard glass tube. It is rated to 1 kbar, which is about three-fold higher than the pressures required for reverse micelle samples in liquid ethane.
The integration of a high pressure valve allows the reverse micelle sample to be transferred to the tube and sealed without the need for maintaining a high pressure line attachment, thus reducing the sample volume required. The cell is easily assembled using two open-end wrenches, and is able to maintain reverse micelle samples at the encapsulation pressure for months.
There is a 1/4"-28 1 kbar fitting for use with 1/16" tubing located on the side of the cell for pressurization prior to closing the valve. The housing is made from 7075 aluminum which has been found to be inert to the organic reagents used in reverse micelle samples.

Support Equipment:
Safety Box: This acrylic walled box provides protection to users while pressurizing cells. It also helps protect the setup while moving samples from the bench to the spectrometer. It also provides a convenient place to store high pressure cells when not in use. The box is open on the bottom.
Bruker and Varian versions are available.
Cell Setup Tool: The high pressure tubes are not permanently fixed in the cell to allow for tubes to be switched and cleaned independently. This benefit requires some care when assembling the high pressure cell. This tool assists in that process by helping to keep the tube axially aligned with the cell during setup which improves seal integrity and lowers the probability of improper fit in the NMR.
Only one is needed for each cell type (Varian or Bruker).
Other Cell Components:
High Pressure Seals (TS01): These form the primary seal between the high pressure tube and manifold. They are single use. One version works for all tube and cell combinations.
Tube Seats (TCSN): This forms a soft mating surface between the tube and the manifold base. It deforms during cell setup to match any uneven contours of the tube head. Typical usage is one for every ten times the cell assembled for reverse micelle applications. It is non-sealing.
